552 Identification of cutaneous Schwann cell subsets based on single cell gene expression analysis

Schwann cells, which surround myelinated/nonmyelinated sensory neurons, play critical roles in transmission, nerve regeneration, and wound healing. Recent studies have revealed the presence of specialized forms called terminal upper dermis peri-follicles. However, how these cells differ from other what they do skin remains unknown. This study aims to establish novel classification cutaneous by using single cell gene expression analysis characterize cells. We crossed Plp-creERT2 mice with ROSA26-loxP-STOP-loxP-tdTomato label all lineages. prepared suspension dorsal back reporter mice, collected sorter, analyzed RNA-sequencing technique. The clusters were roughly divided into two groups: myelinating non-myelinating (nmSCs). further subdivided nmSC found that level Ngfr, a known marker for nmSCs, varied each cluster. Scn7a, another was ubiquitously expressed its inversely correlated Ngfr. Some Scn7a-high nmSCs lacked Sox10, common neural crest-derived detected candidate genes specific differentially cluster evaluated their validity immunohistochemical staining murine skin. Our proposes new landscape heterogeneous populations provides insights may engage pathological processes dermatological conditions.
